Location: Clemson, SC
Job SummaryThe technician will be responsible for creating non‑pathogenic DNA constructs used in single‑molecule biophysics experiments and other duties as assigned. Tasks include assembly of DNA fragments into plasmids using methods such as traditional restriction digest cloning, site‑directed mutagenesis, Gibson assembly, and ligation‑independent cloning. Non‑pathogenic Escherichia coli strains will be transformed with assembled plasmids to produce PCR templates or purified fragments for downstream experiments.
Job Duties- 60% – Essential: Use a variety of molecular cloning techniques to assemble DNA fragments into plasmids. Verify successful assembly by gel electrophoresis and PCR.
- 30% – Essential: Generate purified plasmid fragments via restriction digests or PCR, followed by purification and optional ligation. Manage purification for large‑quantity DNA when required.
- 10% – Essential: Organize storage of DNA and protein aliquots in –20 °C and –80 °C freezer rooms, and maintain an updated database with location information.
- High school diploma and work experience in a research or clinical laboratory setting.
- Bachelor’s degree in a related scientific discipline may substitute for required work experience.
- 0+ years of work experience.
- Associate or Bachelor’s degree with quantitative science coursework.
- 1+ year of experience with molecular biology techniques (restriction digest, ligation, PCR) and microbiological cloning techniques.
- Fundamental Job Knowledge: Working knowledge of concepts, practices, and procedures applicable to the role.

Anticipated Salary Range: $31,800 – $36,000. Salary depends on experience, knowledge, skills, and performance.
Personnel LevelNormal Operations. Required to follow emergency closure directives; not expected to work on‑site during emergencies.
