The project aims to validate, in independent cohorts of autoantibody-positive at-risk of T1D subjects, a model integrating plasma levels of circulating micro
RNAs with clinical variables (glucose/C‑peptide responses, autoantibodies, HbA1c, BMI SDS) to estimate progression to Stage 3 T1D. In parallel, it will dissect the mechanisms underlying this miRNA profile in human immune cells (neutrophils, macrophages, T and B lymphocytes) by modulating key miRNAs (gain/loss-of-function) and analysing expression and activity of mRNA targets involved in immune activation, signalling and inflammation, linking the circulating panel to dysregulated immune pathways driving T1D progression
