Position Summary
Senior Oligonucleotide Scientist (non-viral delivery technical development team). Technical lead for design, synthesis, purification, and analysis of modified oligonucleotides; drive chemical modifications and scale-up process development from discovery through clinical development.
Responsibilities- Lead automated solid-phase synthesis of complex, highly modified oligonucleotides at various scales.
- Develop/optimize purification using preparative RP-HPLC, IEX, SEC, plus TFF and lyophilization.
- Oversee/perform structural elucidation and purity assessments using LC-MS, MALDI-TOF, NMR, and capillary electrophoresis.
- Optimize synthetic routes and cleavage/deprotection methods to improve yields and reduce impurities.
- Evaluate/implement novel chemical modifications and conjugation/delivery strategies (e.g., GalNAc, lipids, peptides) to enhance stability and potency.
- Serve as oligochemistry lead on multidisciplinary teams.
- Supervise/mentor/train junior chemists and technicians; maintain lab notebooks, write SOPs, support regulatory filings.
- Manage inventory of testing articles and critical reagents/phosphoramidites.
- Analyze and communicate results via experimental reports and meetings.
- Ph.D. (Organic/Medicinal or related) with 4.5–8+ years, incl. 2+ years industrial biopharma/biotech OR non-PhD with 12–15 years progressive experience.
- Expertise in phosphoramidite chemistry, protecting groups, and nucleotide modifications (e.g., 2'-OMe, 2'-F, phosphorothioates, locked nucleic acids).
- Hands‑on automated oligonucleotide synthesis (e.g., ÄKTA Oligopilot, Mermade) and preparative purification.
- Strong troubleshooting and interpretation of LC-MS for complex macromolecules.
- ICH and cGMP familiarity (highly desirable).
- Strong problem‑solving; thrive in fast‑paced, matrixed environment.
- Excellent written/verbal communication.
- Proven project leadership and collaboration.
- Base pay range: $145,000–$155,000+ bonus, equity and benefits.
- On‑site (manufacturing/lab‑based role).
