Position Summary
We are seeking a highly motivated Senior Scientist with expertise in LC-MS-based antibody/protein characterization and/or bioanalytical quantitation to support oncology drug discovery. The successful candidate will perform hands‑on analytical research focused on antibody‑drug conjugates (ADCs), antibodies, and small molecules using LC-MS and HPLC technologies. This role partners closely with multidisciplinary teams to develop, optimize, and apply analytical methods that support discovery and early development programs.
Key Responsibilities- Perform analytical characterization and quality control of ADCs, antibodies, and small molecules using LC‑MS and HPLC techniques, including:
Intact mass analysis, de‑glycosylation and reducing LC‑MS, Drug‑to‑antibody ratio (DAR) determination, HPLC methods such as size exclusion chromatography (SEC), hydrophobic interaction chromatography (HIC), and reverse‑phase (RP), peptide mapping (a plus). - Prepare and analyze samples from purified proteins and biological matrices, including plasma, serum, and tissues.
- Identify and characterize ADC metabolites from in vitro and in vivo studies using analytical techniques such as affinity capture, forced release, MS/MS, and SIM‑MS.
- Develop, optimize, validate, and troubleshoot analytical methods to improve performance and resolve technical challenges.
- Operate, maintain, and troubleshoot analytical instrumentation, such as Thermo Vanquish UHPLC‑Orbitrap MS, Waters Acquity UPLC‑SQD MS, Agilent HPLC, and UV‑Vis systems.
- Evaluate and recommend new analytical technologies and instrumentation to expand internal capabilities.
- Document experimental work in electronic laboratory notebooks (ELNs), maintain analytical databases, and prepare technical reports and project summaries.
- Present experimental results and scientific findings to multidisciplinary project teams.
- Contribute to laboratory operations by maintaining shared equipment, supporting laboratory organization, and improving analytical workflows and efficiencies.
- Ph.D. or equivalent degree in Analytical chemistry or a related field with minimum 5 years’ experience, MS degree with 8 years’, or BS degree with 11 years’.
- Industry experience in analytical chemistry within the biopharmaceutical or pharmaceutical industry.
- Hands‑on experience with protein and bioconjugate characterization using LC‑MS, UV‑Vis, and HPLC.
- Experience with protein sample preparation techniques, including de‑glycosylation, reduction, and enzymatic digestions of ADCs.
- Working knowledge of chromatographic methods, including SEC, HIC, and reverse‑phase HPLC.
- Experience operating, maintaining, and troubleshooting LC‑MS and HPLC instrumentation.
- Strong analytical and problem‑solving skills with excellent attention to detail.
- Ability to manage multiple priorities and work independently in a fast‑paced environment.
- Excellent written and verbal communication skills, with the ability to present scientific data to multidisciplinary teams.
- Demonstrated ability to collaborate effectively with cross‑functional teams, including chemists, protein engineers, and biologists.
- Scientific curiosity, a passion for innovation, and enthusiasm for evaluating and implementing emerging analytical technologies.
