Research Associate I – Optical Physiology – In Vivo Indicator Benchmarking

Neural Dynamics is a moonshot research program in systems neuroscience within the Allen Institute. We aim to explain how the brain builds our understanding of the complex world to guide the flexible behaviors that address our biological needs. The answers will be in terms of defined neuron types and circuits interacting across the whole brain and body. We will develop next-generation methods and theories and employ a team-based approach to discovery neuroscience.

Knowledge, data, models, and tools will be widely shared to support the development of therapies for brain disorders.

We are seeking a Research Associate to help systematically benchmark genetically encoded fluorescent indicators used to measure dynamic neural activity in the living brain. In recent years there has been rapid development of such indicators for neuromodulators and neurotransmitters. Systematic and quantitative in vivo comparisons are needed for scientists to select among these indicators and appropriately design their experiments. To this end, we have created protocols to benchmark indicators in vivo using a standardized expression system, in vivo fiber photometry, and physiologically calibrated optogenetic stimulation.

The Research Associate will use these protocols to evaluate state-of-the-art and next-generation fluorescent indicators for brain research. The ideal candidate will have a background in biological sciences with a passion for collaborative work in a fast-paced team environment. Prior experience is not required in all the above techniques; appropriate training will be provided.

Experimental approaches and responsibilities will involve processing mouse brain tissue, histological staining, stereotaxic surgery, and fiber photometry methods. The candidate will be responsible for executing laboratory tasks and/or in vivo experiments following Standard Operating Procedures (SOPs). Good judgment and problem-solving skills are required for recognizing process deviations, participating in troubleshooting, and operation of complex laboratory instrumentation. This role is expected to comply with regulatory requirements, adhere to SOPs, initiate individual experiments, and to work efficiently in a team environment.

Essential Functions

  • Independently carry out experiments involving fiber photometry and optogenetics
  • Train mice to perform simple behaviors in an experimental setting
  • Perform stereotaxic survival surgeries in mice
  • Section, stain and mount tissue for high resolution confocal microscopy
  • Follow standardized operating procedures, good laboratory practices, and regulatory requirements
  • Maintain clear and accurate communication within and between teams
  • Coordinate with teammates to prioritize tasks and manage project timelines
