We are seeking a mid-to-senior level laboratory technician specializing in CRISPR screening to independently plan and execute pooled CRISPR loss- and gain-of-function screens in mammalian cell lines. A successful candidate will own the full experimental workflow end to end, including sgRNA library design and construction, high-titer lentiviral production, low-MOI transduction, antibiotic selection, FACS-based cell sorting, cell culture, genomic DNA isolation, and sgRNA library preparation for next-generation sequencing.
Ideal candidates are meticulous, safety-conscious bench scientists who thrive on troubleshooting long, multi-stage workflows and take full ownership of screen quality from library design through sequencing handoff.
This position is in New York (NY) and requires full-time, in-person work in a laboratory setting.
Responsibilities- Design, construct, and validate sgRNA expression libraries for pooled and/or arrayed screens, including guide selection, development of cloning strategies, and plasmid library QC
- Produce, concentrate, and titer high-quality lentivirus preparations at scale to support genome-wide or focused sub-pool screens
- Execute low-MOI transductions and antibiotic selection while preserving full library representation
- Perform FACS-based sorting for phenotypic screen readouts, including gating strategy design and post-sort recovery
- Maintain mammalian cell culture across all stages of a screen, from pilot to full scale
- Isolate high-quality genomic DNA and prepare barcoded sgRNA libraries for next-generation sequencing
- Track coverage, representation, and QC metrics at every stage and proactively flag deviations
- Author and maintain detailed protocols, records, and SOPs to ensure reproducibility across screens
- Coordinate with sequencing team, vendors, and bioinformatics collaborators on sample handoff and specifications
- B.S. in Molecular Biology, Cell Biology, Biochemistry, Genetics, or a related field (M.S. a plus) 3+ years (mid-level) or 6+ years (senior) of full-time, hands-on mammalian cell culture and molecular biology experience
- 1+ years (mid-level) or 3+ years (senior) of direct, hands-on experience with lentiviral production, titering, and transduction
- 1+ years (mid-level) or 3+ years (senior) of experience executing all components of a pooled CRISPR screen (plasmid library construction, virus production, selection, and sequencing library prep)
- 1+ years (mid-level) or 3+ years (senior) of experience preparing samples for flow cytometry/FACS, plus a working understanding of sorting and gating principles
- 1+ years of proficiency in genomic DNA extraction, PCR, and NGS library preparation techniques
- Meticulous attention to detail and rigorous documentation habits
- Strong troubleshooting skills across long, multi-step, interdependent workflows
- Ability to work independently and manage multi-week experimental timelines
- Standout Experiences Independent ownership of a full pooled CRISPR screen from library design through sequencing handoff
- Experience with genome-wide screening libraries (e.g., Brunello, GeCKO, Cellecta)
- Familiarity with screen-analysis pipelines (e.g., MAGeCK, BAGEL, drugZ) and ability to interpret QC and hit-calling outputs
- Hands-on operation of a cell sorter (e.g., BD FACSAria-class instrument)
- Experience with CRISPR screen variants beyond knockout (CRISPRi, CRISPRa, base editing)
- Publication or major technical contribution to a CRISPR screening study
- Basic scripting ability (Python or R) to support screen QC and data handoff
