Job OverviewWe seek a Principal Scientist with 5+ years of biologics antibody sequencing with experience working on 3 or more antibody drugs or biosimilars at top pharma (e.g., Abb Vie, Amgen, Genentech) and proven expertise in making and submitting FDA antibody approval data packages
. This role supports diagnostics and therapeutics manufacturing with advanced analytical skills.
- Sequence antibody heavy/light chains using mass spectrometry (LC-MS/MS, Orbitrap, QTOF) for 100% coverage.
- Perform N-to-C and C-to-N sequencing, resolving leucine/isoleucine and PTMs (e.g., glycosylation).
- Characterize antibodies (disulfide mapping, glycan profiling, heavy/light chain validation) for diagnostic/therapeutic applications.
- Optimize ELISA assays for quantification, affinity, and function.
- Conduct glycan analysis via HILIC-MS or released glycan workflows.
- Analyze MS data with Byonic, Protein Metrics, or PEAKS AB (intact mass ±2 Da).
- Maintain/troubleshoot MS and HPLC/UHPLC systems.
- Collaborate to deliver results for diagnostics and therapeutics
, including making and submitting FDA antibody approval data packages
. - Document experiments for regulatory submissions.
- Education: Ph.D./M.S. in Biochemistry, Analytical Chemistry, or related field (B.S. with 7+ years considered).
- Required Experience: 3+ years of biologics antibody sequencing experience with 3 or more antibody drugs or biosimilars at top pharma (e.g., Abb Vie, Amgen, Genentech).
- Proven expertise in making and submitting FDA antibody approval data packages.
- Expertise in N-to-C/C-to-N sequencing (top-down/bottom-up MS).
- Skilled in PTM analysis, glycan profiling, ELISA development.
- Proficient in MS data tools (Byonic, Max Quant, PEAKS AB).
- Preferred
Skills:
Experience with Maurice, CE-SDS, or iCIEF for FDA characterization. - Knowledge of FDA/ICH validation guidelines.
- Familiarity with high-throughput proteomics/automation.
- Skills: Strong MS/MS data analysis and problem-solving.
- Detail-oriented, multi-tasking, collaborative.
