Myoventive is a precision-cardiology therapeutics company developing next-generation oligonucleotide-based medicines to transform the treatment of genetic heart disease. We are seeking a highly skilled and hands-on Senior to Principal Scientist to join our Protein Science team.
This role will focus on developing and executing antibody-oligonucleotide conjugate (AOC) production processes at in vivo-enabling scale. The successful candidate will optimize conjugation and purification workflows and deliver reproducible, high-quality AOC material for pharmacology and PK/PD studies.
This position is ideal for a scientist who thrives at the bench, enjoys solving conjugation, purification, and scale-up challenges, and wants to apply deep bioconjugation and process development expertise to a novel therapeutic modality in a fast-moving biotech environment. The role is expected to deliver dosing-ready AOC material on study timelines while establishing robust and repeatable production workflows.
Key Responsibilities- Develop and execute protein-oligonucleotide conjugation processes at in vivo-enabling scale, including approximately 100 mg batches for preclinical studies, with further scale-up to support planned non-human primate studies.
- Optimize conjugation conditions to improve efficiency, yield, product quality, and batch-to-batch reproducibility.
- Identify and resolve process challenges including aggregation, conjugation efficiency, reagent losses, solubility limitations, and product heterogeneity.
- Optimize reagent utilization and recovery, particularly for supply-limiting oligonucleotide and linker inputs.
- Develop and execute preparative purification workflows for AOCs using appropriate chromatography and filtration approaches.
- Develop buffer exchange and concentration workflows to support dosing requirements and downstream studies.
- Plan and execute AOC production campaigns against study timelines, coordinating antibody supply and externally sourced oligonucleotide and linker inputs.
- Establish process controls and fit-for-purpose QC criteria to support consistent AOC production.
- Work closely with Protein Analytics to assess identity, purity, aggregation, free oligonucleotide, concentration, and other relevant product attributes.
- Evaluate material stability and handling requirements, including storage, freeze-thaw, shipping, and dosing conditions.
- Develop and document workflows, batch records, and SOPs to support reproducible execution and future external process transfer as needed.
- Work closely with colleagues to align conjugate design, analytical characterization, material requirements, formulation, and study timelines.
- Communicate clearly and proactively on timelines, technical risks, and material requirements.
- Use process and scale-up learnings to inform molecule design and contribute to advancement of the AOC platform and related intellectual property.
Required:
- Ph.D. in Chemistry, Biochemistry, Chemical Engineering, Chemical Biology, or related field.
- 5+ years of relevant industry experience for Senior Scientist; 8+ years for Principal Scientist.
- Strong hands-on experience with bioconjugation chemistry, including development and optimization of conjugation and purification workflows.
- Demonstrated hands-on experience producing and purifying bioconjugates at approximately 100 mg to gram scale for in vivo studies, including preparative chromatography.
- Working knowledge of analytical methods used to characterize protein conjugates, such as SEC, IEX/HIC, LC-MS, and CE-SDS.
- Experience independently designing experiments and driving technical problems to resolution under study timelines.
- Prior experience with AOCs, ADCs, or related bioconjugate modalities.
- Experience with oligonucleotide payloads (ASO, siRNA, PMO), including handling, purification, and stability considerations.
- Experience with scale-up purification, buffer exchange, and concentration workflows for protein or bioconjugate therapeutics.
